Services in proteomics

Two dimensional electrophoresis

Two dimensional gel electrophoresis
Image analysis using Image Master 2D Platinum software (min. 1hr)

Molecular Weight Determination

Direct infusion into electrospray MS/MS (MS spectrum)
Direct infusion into electrospray MS/MS, selecting precursor max. 3 ions, acquiring of MS/MS spectra

Protein identification

Enzymatic digestion
Capillary UPLC separation followed by MS/MS detection (data directed analysis)  (Note: at least 4 analyses are necessary for a reasonable result)
Database searching

Sample:
Depending on the mass of the protein molecule, 1-5 pmol of sample is required. More sample material is necessary if the protein identification is requested. We will appreciate it very much having from you all information concerning the sample to be analyzed (e.g. what kinds of buffers, detergents, etc. are in sample). In addition, we will be pleased knowing from you what data are preferentially expected from the analysis. 

Method and Instrument:
Samples are analyzed on a Waters Q-tof Premier mass spectrometry instrument. The Q-Tof Premier™ is a quadrupole, orthogonal acceleration time-of-flight tandem mass spectrometer that builds on the success of the seminal Q-Tof instrument range by offering enhanced levels of flexibility, sensitivity, specificity and speed of acquisition.

Compositional and linkage analysis of carbohydrate polymers from biological materials

One dimensional SDS-PAGE

Separation of polysaccharide/protein complex mixtures on HPLC

Sample:
For GC or GC/MS analysis at least 0.5 mg of sample is required. In the case of methylation linkage analysis about 1 mg of sample is necessary to obtain reasonable results.  We will appreciate it very much having from you all information concerning the sample to be analyzed (e.g. what kinds of buffers, detergents, etc. are in sample). In addition, we will be pleased knowing from you which data are preferentially expected from the analysis. 

Methods and Instruments:
GC and GC/MS are performed on a Shimadzu Gas Chromatograph 17A and a Shimadzu Gas Chromatograph Mass Spectrometer GCMS-QP5000. 
HPLC includes separations/purifications of various polysaccharide/protein complex mixtures recorded with RI detector.
